En amerikansk författare vid namn Peter Neal (Anthony Franciosa) är i Rom för att göra reklam för sin nya bok. Han får snart reda på att en seriemördare härjar, och att mördaren tar inspiration från olika kapitel i hans bok "Tenebrae". Detta märks inte minst när mördaren<br/>har rivit ut sidor ur boken och lagt på mordplatsen, eller kört in i munnen på offret.<br/>Det visar sig senare att det handlar om FLERA mördare och polisen får en "jolly good time" med att komma på vilka det kan vara.

När den ökände förbrytaren Ben Wade slutligen grips ser den fattige boskapsuppfödaren Dan Evans en möjlighet att rädda sin torkdrabbade gård. För 200 dollar erbjuder sig den annars så fredlige bonden att eskortera den tillfångatagne brottslingen till den närliggande staden Contention, där de ska ta 3:10-tåget till Yuma-fängelset. Färden till tågstationen är dock farlig och Wades hänsynslösa gäng är dem hack i häl med avsikt att rädda sin ledare.
